This write up will account for all your time you have spent on researching your soil organisms. It will begin with you bringing in your soil samples, and end with your bacterial identification of your chosen isolate.
Introduction:
No more than 4-6 sentences. Briefly, what was the goal of the experiment?
Example:
The purpose of these experiments was to isolate soil bacteria in hopes of discovering a novel antibiotic producer, and identifying isolated bacteria to a genus and species level. This was completed over a number of weeks and utilized multiple microbial inoculation techniques.
Methods:
For each of the labs, you will succinctly describe the lab process. Basically, you will walk the reader through the process in case they wanted to recreate what you have done. To be included in this section are the following labs:
Soil sample collection
Serial dilution and spread plating of your sample
Making two master plates from your dilutions
Patching your isolates against the ESKAPE-safe relatives
Inoculation of the biochemical tests
Identification of your isolate
Example:
This soil sample was collected at 5:35pm on September 22nd. The temperature of the air was approximately 72°F at time of collection. The sample was collected at GPS coordinates 37.665091795594584, -121.03675682886293 which corresponds to the edge of Sierra parking lot at Modesto Junior College, west campus. The top 3 inches of soil were removed prior to sample collection. Sample was a mix of sand and loam with moderate water content. A small scoopula was used to collect the sample in to a 20mL conical vial with screw top lid. The tube was filled about half way with soil to ensure enough to use for dilutions. The sample was stored overnight at room temperature and brought to campus the next day for further study.
You see in the above example, enough information was given that the reader not only knows where the sample is from, but also could recreate the collection using the provided information. That is the goal for each methods section. Can the reader understand what you did, and can the reader recreate it? Do not just copy/paste the materials and procedures from the lab printouts. Use full sentences and put in to your own words.
Results from the ESKAPE-safe relative patches and bacterial identification:
For the ESKAPE-safe relative patches, take a photo of your plates. Describe any isolates that seem to have produced an antibiotic that was inhibitory to the ESKAPE-safe relative. Attach the photo to your document here.
For the bacterial identification, this must be in a TABLE format!!!! The table must have a table title and a legend. Include ALL results. (4 plates and 3 tubes, catalase and Gram stain results.)

Discussion:
Here is where you describe the results of the whole activity. If you were able to positively identify your isolate, here is where the name will go (using appropriate nomenclature). Did everything run smoothly or did you have snags? (Things like fungal contamination, lack of isolated colonies, inability to identify the organism.) For all your biochemical tests, describe your organism’s reactions in them. Describe what the tests test for and how you knew you had the result you did. For example, if you logged a + catalase test you would say something like the following; “Isolate #16 (or whatever) was + for catalase. After the addition of hydrogen peroxide, bubbles were seen. Catalase is an enzyme that converts toxic peroxides in to no toxic products.
The whole document should be in the neighborhood of 5 pages including your tables and data sheet.
